Building 340B Resilience: 340B Coalition Session Recap

Reflections from 340B Coalition's Summer Conference and our session “Building 340B Resilience: Preparing for Regulatory Change with Automation” hosted on July 23, 2025.
At this year’s 340B Coalition Summer Conference, our Founder & CEO, Joy Liu, had the privilege of speaking with an incredible group of pharmacy leaders about a topic we’re deeply passionate about: how workflow automation and other technology innovations can help healthcare teams build resilience, especially given so many legislative changes impacting 340B teams.
Check out the key takeaways from her session below. And if operational efficiency and modernization is top of mind for your team, grab time with our team for a demo of what we’re building here at Plenful.
Why 340B Needs Innovation Now
Joy spent her career inside healthcare operations, from specialty pharmacy to enterprise health systems, and knows firsthand how challenging that work is. Today, regulatory changes like the Inflation Reduction Act (IRA) and upcoming Maximum Fair Price (MFP) rules are raising the stakes. Teams are being asked to do more, track more, and report more, all while managing tighter margins.
But the reality is that most 340B workflows are still heavily manual. A lot of information is passed from stakeholder to stakeholder via flat files. People’s desks are littered with Post-It notes in an effort to stay organized. Whether it’s auditing claims, reconciling referrals, or managing reporting for ESP and state requirements, these processes eat up hours. And even then, critical savings opportunities are still missed. This is where workflow automations can be transformative.
Turning Complexity into Opportunity
At Plenful, we’ve built an agentic operating system, where configurable rules-based automations fit within existing workflows to work alongside operational teams. Not as a “black box,” but as a transparent partner. We’ve seen Covered Entities (CEs) recapture millions of dollars by using our platform to uncover 340B savings, prioritize high-value claims, and surface referral opportunities that would otherwise go unseen.
Joy shared one story at the conference about a large academic medical center in the Midwest. By leveraging Plenful to audit their 340B claims marked ineligible by their TPA, our platform was able to locate where errors were made and flag the claims that actually were eligible. As a result, they uncovered $2 million in missed savings in the first month alone. That’s the kind of bottom-line impact our agentic operating system can have when it’s designed with healthcare workflows in mind.
But beyond revenue, our platform also plays a huge role in 340B compliance. Rather than relying on spot-checks or sampling, Plenful allows teams to review every claim, flagging anomalies early and maintaining audit readiness with confidence.
Scaling Without Adding Headcount
One of the biggest themes our team heard at 340B Coalition was this: “We need to scale, but we can’t afford to add headcount.” This is something we’re thinking about every day. Staffing shortages, administrative overload, and competing priorities make it nearly impossible for pharmacy teams to do everything manually.
Automation solves this challenge by handling the repetitive administrative work, like benefits verification, prior authorization, and claims reconciliation, while giving staff time to focus on high-value, strategic tasks. With our 340B Audit & Optimization Management suite, powered by a configurable rules engine, Plenful helps teams eliminate over 95% of the work.
Building Resilient, Agentic Operations
What excites us most about the future of 340B is the idea of transforming siloed expertise into scalable infrastructure. That’s the promise of an agentic operating system like what we’re building at Plenful. Instead of relying on a handful of experts to keep everything running, we’ve built our platform and customer configurations to amplify the care team’s knowledge and to automate their processes in a way that scales across the organization.
This means expansive, configurable dashboards that show you exactly where you stand, highlighting compliance metrics, turnaround times, and revenue capture all in one place. It also means being ready for what’s coming, from MFP-driven reimbursement changes in 2026 to new reporting requirements for Covered Entities participating in 340B.
Practical Advice for Getting Started
In Joy’s session, she encouraged attendees to focus on three best practices when adopting workflow automation technologies like Plenful to streamline manual work:
- Start with high-impact use cases. Pick a workflow where automation will deliver a clear ROI, like 340B missed opportunities or across your intake authorization journey.
- Insist on configurability. The technology you choose needs to be highly-configurable to support your unique system’s processes, to integrate with your disparate data sources, and to report on the KPIs that matter most to you.
- Bring your team along. Workflow automation technology works best when it’s designed to complement your staff’s expertise, not replace it. Involving them early builds trust and facilitates user adoption once the tool is implemented.
The Time to Act Is Now
Between the legislative changes coming from the IRA and increasing scrutiny on 340B programs, now is the time to invest in operational resilience. Automation is no longer a nice-to-have; is a strategic enabler for capturing savings and safeguarding compliance in an evolving landscape. And the 340B leaders who embrace it now will be better equipped to thrive as complexity grows.
